There are several methods for checking for sensitivity of acetylcholine esterase in insects using a single insect. It comprises the steps, in any order, of homogenizing an only insect in aqueous neutral to acid pH phosphate buffer. It produces a result of acetylcholine iodide in neutral to acid pH phosphate buffered water-miscible organic solvent. And then by producing a solution of 5,5-dithio-bis in the buffer and producing a solution of propoxur in the buffer by dropping the insect homogenization into the wells of an assay plate. Drop the acetylcholine iodide and 5, 5-dithio-bis, solutions into one of the well and 5, 5-dithio-bis propoxur solutions into the other well. Now check for a difference in the yellow colouration between the samples in the two sets of cells. The invention also provides a test kit for formative susceptibility of acetylcholine esterase in insects using a single insect comprising a pot containing separate containers.

Why it is needed: Chemical insecticides keep on as the most important agents for the control of vector-borne diseases like malaria, dengue, filartasis etc. Countless deaths from these diseases have been secured since the beginning of the first synthetic insecticide, DDT. Prolonged use of insecticides has though, induced the development of fighting in the vectors, primarily insects that carry diseases. The use test kits of short shelf life and need skilled manpower to conduct the tests and interpret the test results are very fruitful. As a result, very often little is known of the susceptibility status of many insect vectors which tends to affect the outcomes of control programs.

Advantages: The growth of rapid field test kits would be a key breakthrough in the control of vector-borne diseases. In common, the biochemical basis of confrontation is due to one of three mechanisms like

1. Increased level of non-specific esterase's targeted against organophosphates and carbamates.

2. elevated action of mixed-function oxidases against pyretroids

3. Non-susceptibility of acetylcholine esterase beside organophosphates and carbamates.

According to the creation the test kit comprises a container containing detach containers of a neutral to acid pH phosphate buffer solution, acetylcholine iodide, 5,5-dithio-bis-2-nitrobenzoic acid, propoxur and a water-miscible organic solvent.
